Model Update of Signal Transduction Pathways in Hepatocytes based upon Sensitivity Analysis

his work presents sensitivity analysis of a model describing JAK/STAT and MAPK signaling to quantify the contribution of individual parameters to signal transduction in hepatocytes stimulated by interleukin-6 (IL-6). Results from the analysis are used as a guideline for which parts of the model benefit from further model refinement. Based upon this analysis an updated version of the model is derived and validated in experiments involving a green fluorescent protein (GFP) reporter system. The model predictions are in excellent agreement with the experimental results.
